【摘要】:Sulfate-reducingbacteriaarewidelydistributedinmarineandfreshwatermudswhich,inconsequence,frequentlyareladenwiththehydrogensulfideproducedbytheseorganismsduringdissimilatorysulfatereduction.IthasbeenreportedthatDesulfovibriocanformasmuchas10gofsulfideperlitreduringactivemultiplication.Sulfate-reducingbacteriacancausetheexternalorinternalcorrosionofwaterorwastewaterpipelinesandpipelinesforpetroleumandnaturalgas.Theformationofgalvaniccellsbymassivegrowthofsulfate-reducingbacteriaundersuitableconditionsmakesthecorrosionmuchworsethanjusttheeffectofthehydrogensulfideonthemetalorconcrete.1.1Thesetestmethodscovertheprocedureforthedetectionandenumerationbythemostprobablenumber(MPN)techniqueofsulfate-reducingbacteriainwaterorwater-formeddeposits.1.2Twomediapreparationsareprovided.MediumAwhichispreparedwithreagentgradewater,andMediumBwhichispreparedusingthewatertobesampledasthewatersource.MediumBisofferedforthosespecialconditionswheresulfate-reducingbacterialstrainshaveadaptedtoatypicalnon-freshwaterenvironment.1.3Fortheisolationandenumerationofthermophilicsulfate-reducingbacteriaencounteredinwatersassociatedwithoilandgasproduction,allbroths,dilutionblanks,andincubationsmustbemaintainedattemperaturesofatleast45176;Candpreferablywithin5176;Catthesampletemperature.1.4Thesensitivityofthesetestmethodscanbeincreasedbypurgingthedilutionblanksandtubesofmediawithnitrogenimmediatelypriortouse.1.5TheanalystshouldbeawarethatadequatecollaborativedataforprecisionandbiasstatementsasrequiredbyPracticeD2777arenotprovided.SeeSection11fordetails.1.6Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
